Pharmacological Properties |
|
The synergistic combination of Sulfadoxine and pyrimethamine. These ingredients act by blocking two enzymes involved in biosynthesis of folinic acid in parasites. Malagon (Sulfadoxine & Pyrimethamine) is also sensitive and most effective to resistant organisms, to conventional antimalarials like Chloroquine and its derivatives. The incidence of resistance with Malagon is rare and negligible. |

Malagon is absorbed orally and excreted mainly by kidney. Following a single dose administration of Malagon (Sulfadoxine & Pyrimethamine). Sulfadoxine peak plasma concentration 51 to 76 mcg/ ml were achieved in 2.5 to 6 hours and Pyrimethamine peak plasma concentration 0.13 to 0.4 mcg/ml were achieved in 1.5 to 8 hours. The approximate elimination half life of Sulfadoxine ranges from 100 to 231 hours with a mean of 169 hours where as pyrimethamine half life ranges from 54 to 148 hours with a mean of 111 hours. |

Indications |
|
Malagon is indicated for the treatment and prophylaxis of malaria caused by plasmodium falciparum, plasmodium vivax, plasmodium ovale and plasmodium malariae. Malagon is also effective for chloroquine resistant malaria. |

Malagon has also been found effective in infection caused by toxoplasma gondii. |
|
Malagon is also effective for the prophylaxis of pneumonia due to pneumocystis carinii. |

Contraindications |
|
Malagon (Sulfadoxine + Pyrimethamine) is contraindicated in patients with known hypersensitivity to Sulfadoxine or Pyrimethamine. It should be avoided as safety is not established in pregnancy, lactation and premature babies. |
Side Effects |
|
Generally very well tolerated. Rarely reported in negligible cases of nausea, with or without vomiting, skin rashes. In rare cases headache, fatigue, fever is also reported. |
Precautions |
|
Malagon should be discontinued if skin rashes appear. Oral intake of fluid should be increased. To be given with caution in patients with hepatic damage. |
